The striker, who joined the club from Oldbury two weeks ago, made it three goals in two games since joining Spartak.
Brookes scored in the 20th minute to put Spartak ahead.
Vale were then reduced to ten men when Mal Powers was sent off for two yellow cards in the 40th minute. Just before half time Castle Vale had a second player dismissed for swearing at the assistant referee.
In the second half Brookes bagged his third, before winger Sean Williams made it 3-0.
